We have a 4 node VMware-Nutanix cluster it has the following limitations
- vCenter server is running on the same cluster and is not managing any other cluster
- Prism Central VM is deployed on the same cluster
- Nutanix Files is deployed on the cluster

I am planning to upgrade Nutanix Files to version 5.0 first.
