This is a new cluster or it is just the first time you are trying to create a VM from Prism?
Anyway, you need a vSphere cluster with your Nutanix nodes on the cluster, the vCenter registered on Prism and vSphere cluster with DRS enabled. Please refer to the following documentation on how to configure your vCenter according with Nutanix best practices: https://portal.nutanix.com/#/page/docs/details?targetId=vSphere-Admin6-AOS-v510:vSphere-Admin6-AOS-v510
